About this compound
AICAR (5-Aminoimidazole-4-carboxamide ribonucleotide, also known as Acadesine) is a cell-permeable nucleoside analogue and AMP-kinase (AMPK) activator. Upon cellular uptake, AICAR is phosphorylated to ZMP (AICA ribonucleotide monophosphate), a structural mimic of AMP that directly activates AMPK by binding to its regulatory γ subunit. AMPK is a master energy-sensing kinase that coordinates cellular responses to metabolic stress, regulating glucose uptake, fatty acid oxidation, mitochondrial biogenesis, and protein synthesis. AICAR has been extensively used in preclinical research as a pharmacological tool to study AMPK-dependent pathways, particularly in the context of metabolic regulation, exercise mimicry, cardioprotection, and cellular energy homeostasis.

- Rodent exercise studies demonstrated that AICAR administration activated skeletal muscle AMPK and upregulated PGC-1α-dependent gene expression, producing transcriptional adaptations consistent with endurance training in sedentary animals (Narkar et al., 2008)
